Description:
The eB114 antibody reacts with the RASSF1A isoform of the tumor suppressor gene RASSF1. RASSF1A contains a Ras Association Domain (RA) and binds Ras in a GTP-dependent manner to effect apoptosis. The RASSF1A isoform undergoes epigenetic inactivation in various human primary tumors, associated with hypermethylation of CpG islands in its promoter region. This antibody was generated against a synthetic peptide corresponding to the splice junction region of RASSF1A.

Applications Reported:
The eB114 antibody has been reported for use in immunoprecipitation and immunoblotting (WB).

Applications Tested:
The eB114-10H1 antibody has been tested by Western blotting and can be used at approximately 1–10 µg/mL. RASSF1A is detected as a 45 kDa band in Western blotting of HeLa cell extract. It is recommended that the reagent be carefully titrated for optimal performance.

Purity: Greater than 90%, as determined by SDS-PAGE.
Aggregation: Less than 10%, as determined by HPLC.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

This gene encodes a protein similar to RAS effector proteins. Loss or altered expression of this gene has been associated with the pathogenesis of various cancers, suggesting its tumor suppressor function. Inactivation of this gene correlates with hypermethylation of its CpG-island promoter region. The encoded protein interacts with DNA repair protein XPA, inhibits cyclin D1 accumulation, and induces cell cycle arrest. Seven alternatively spliced transcript variants encoding distinct isoforms have been reported.
