Product Specific Information

The B2D monoclonal antibody reacts with the mouse RORgamma(t) protein, a member of the retinoic acid-related orphan receptor (ROR) family, which also includes RORalpha and RORbeta. These ligand-dependent transcription factors regulate multiple physiological processes.

RORgamma is expressed in liver, lung, muscle, heart, and kidney. An alternative transcription form, RORgammat, is expressed exclusively in lymphoid cells such as CD4+CD8+ double-positive thymocytes, Th17 cells, and lymphoid tissue inducer (Lti) cells.

The RORgammat isoform differs from RORgamma by three unique amino acids at the amino terminus. Therefore, the B2D antibody reacts with both RORgamma and RORgammat isoforms.

Target Information

ROR gamma (NR1 Thyroid Hormone-Like receptor) is involved in thymopoiesis, bone metabolism, T-cell apoptosis, and lymphoid organogenesis. It promotes thymocyte survival via activation of the antiapoptotic protein Bcl-x(L) and is essential for lymph node and Peyer’s patch development.

The thymus-specific isoform RORgamma(t) inhibits Fas ligand expression and cytokine secretion in immature thymocytes. ROR gamma binds as a monomer to response elements containing a GGTCA core motif preceded by a 6 bp AT-rich sequence.

Expression has been documented in mouse thymus, adipose, bone, skeletal muscle, liver, and kidney, and in human skeletal muscle. ESTs have been isolated from human tissues including cancerous and normal head/neck, germ cell, prostate, colon, lung, liver/spleen, and pancreas.

ROR gammat (RORgt) is an isoform of RORC.
