Description:
The 8D4-8 antibody reacts with human interleukin-4 (IL-4), a 15–19 kDa cytokine secreted by Th2 cells.

Applications Reported:
The 8D4-8 antibody has been reported for use in capture of human IL-4 by ELISA and in intracellular staining for flow cytometric analysis.

Applications Tested:

• Tested as a capture antibody in sandwich ELISA for analysis of human IL-4.
• Used with biotinylated MP4-25D2 antibody (Product #13-7048-81) for detection and recombinant human IL-4 (Product #14-8049-80) as standard.
• Suitable range for ELISA capture: 1–4 µg/mL.
• Include a standard curve with recombinant IL-4 ranging 500 pg/mL – 4 pg/mL.
• Fluorochrome-conjugated 8D4-8 antibody tested for intracellular staining in flow cytometry at ≤0.5 µg/test.
• A test = antibody amount staining a sample in 100 µL volume (10⁵–10⁸ cells/test).
• Antibody titration recommended for optimal performance.

Target Information

Interleukin-4 (IL-4) is a pleiotropic, immune-modulatory cytokine produced by activated T cells, serving as a ligand for the IL-4 receptor.
IL-4 and IL-13 share overlapping functions via receptor binding. STAT6 plays a key role in IL-4-mediated immune regulation.
The IL-4 gene is part of a cytokine cluster on chromosome 5q (with IL-3, IL-5, IL-13, CSF2).
IL-4 is closely located near IL-13 and IL-5, coordinated by long-range regulatory elements (~120 kb).
IL-4 delta 2 is a known splice variant.
