Product Specific Information

Description:
The MIH5 monoclonal antibody reacts with mouse B7-H1 (PD-L1), a B7 family member with a predicted molecular weight of ~40 kDa. B7-H1 is expressed on T, B, NK, and dendritic cells, and serves as a ligand for PD-1. The PD-1/PD-L1 interaction inhibits T and B cell responses. MIH5 is a blocking antibody.

Applications Tested:

  • Tested by flow cytometric analysis of mouse splenocytes.
  • Recommended ≤0.5 µg/test (in 100 µL final volume).
  • Cell number: 10⁵–10⁸ cells/test (empirically determined).
  • Optimize titration for best assay performance.

Storage and Handling:

  • Use in a sterile environment.
  • 0.2 µm post-manufacturing filtered.

Target Information

Programmed death receptor ligand 1 (PD-L1, B7-H1) is a B7 family member that binds to PD-1, a receptor that negatively regulates T-cell receptor (TCR) signaling. PD-L1 engagement decreases TCR-mediated proliferation and cytokine production. PD-L1 is expressed abundantly in murine and human cancers and is upregulated by IFN-γ, suggesting its role in tumor immune evasion.
