Target Information
LCK는 Src-family tyrosine kinase의 구성원으로, T-cell receptor (TCR) 복합체를 통한 신호 전달에 필수적인 역할을 합니다.
TCR이 활성화되면, LCK는 zeta 서브유닛의 ITAM 모티프를 인산화하여 ZAP70의 SH2 도메인 결합 부위를 형성합니다.
LCK는 또한 ZAP70을 인산화 및 활성화하여 LAT 어댑터 단백질을 통한 후속 신호 전달 플랫폼을 생성합니다.
대부분의 LCK는 세포막에 존재하지만, 일부는 Golgi apparatus에 위치하여 약한 TCR 자극 조건에서 Raf 활성화에 관여할 수 있습니다.
LCK는 N-말단에 myristylation 및 palmitylation 부위를 포함하며, PTK, SH2, SH3 도메인을 가지고 있어 다양한 단백질-단백질 상호작용을 매개합니다.
또한 LCK는 여러 자극에 의해 유도되는 apoptosis 조절에 관여하지만, death receptor에 의해 유도되는 apoptosis에는 관여하지 않습니다.
LCK와 관련된 질환으로는 Immunodeficiency 22 및 CD45 deficiency가 있으며, LCK 유전자의 다양한 splice variant가 보고되었습니다.
