Product Specific Information

The M17/4 monoclonal antibody reacts with mouse CD11a, the 180 kDa integrin alpha L, also known as LFA-1 alpha chain. LFA-1, formed by non-covalent association of CD11a with CD18, is an adhesion molecule involved in lymphocyte and granulocyte function. Ligands include CD54 (ICAM-1), CD102 (ICAM-2), and CD50 (ICAM-3). CD11a is expressed on all leukocytes.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested with mouse bone marrow cells at ≤1 µg/test (100 µL final volume). Optimal titration recommended.

Super Bright 600 Characteristics:

• Tandem dye, excited with 405 nm violet laser, emits at 600 nm
• Recommended filter: 610/20 bandpass
• Sensitive to light; protect from photo-induced oxidation
• Use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize polymer interactions

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) with minimal brightness loss. Clone-specific performance should be determined empirically.

Target Information

CD11a (LFA-1 alpha, ITGAL) forms LFA-1 complex with CD18. It mediates leukocyte adhesion via ICAM-1, -2, -3 and contributes to lymphocyte signaling. Expressed on leukocyte membranes, CD11a transitions from low- to high-affinity conformations upon stimulation, enabling tight ICAM binding. It is involved in immune cell interactions, differentiation, proliferation, and T-cell effector functions.
Blocking CD11a is a therapeutic target for inflammatory diseases; e.g., Efalizumab (Raptiva) inhibits T-cell migration. Simvastatin binding to CD11a modulates immune activity.
Associated diseases include benign cephalic histiocytosis and leukocyte adhesion deficiency.
