Product Specific Information

The eBioSN5c monoclonal antibody recognizes human CD10 (CALLA, NEP, enkephalinase, Neprilysin), a 100 kDa type II cell surface glycoprotein originally identified on acute lymphoblastic leukemias (ALLs). CD10 is a Zn²⁺-dependent metallopeptidase acting on substrates such as endothelin, glucagon, gastrin, neurotensin, and bradykinin. It regulates chemotactic and inflammatory processes involving neutrophils and is expressed on B cells, granulocytes, and bone marrow stromal cells.

Applications Reported:
This antibody is reported for use in flow cytometric analysis.

Applications Tested:
Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (0.25 µg) per test in a final volume of 100 µL. Cell number may range from 10⁵ to 10⁸ cells/test.

Fluorochrome Information:
Super Bright 600 is a tandem dye excited with the violet laser line (405 nm) and emits at 600 nm. Recommended filter: 610/20 bandpass. Ensure instrument compatibility.

Staining Buffer Recommendation:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Protect vial and stained samples from light to prevent photo-induced oxidation.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

CD10 (Common Acute Lymphocytic Leukemia Antigen, CALLA) is a cell surface enzyme with neutral metalloendopeptidase activity that inactivates biologically active peptides. It is expressed on lymphoblastic, Burkitt’s, and follicular germinal center lymphomas, immature B cells, CML cells, breast myoepithelial cells, bile canaliculi, fibroblasts, and epithelial cells of kidney and gut. CD10 cleaves peptides at the amino side of hydrophobic residues and inactivates peptide hormones including glucagon, enkephalins, substance P, neurotensin, oxytocin, and bradykinin. It is a 100 kDa type II transmembrane glycoprotein. The 5′ untranslated region of the CD10 gene is alternatively spliced into four mRNA transcripts.

Diseases associated with CD10 dysfunction include spinocerebellar ataxia 43 and Charcot-Marie-Tooth disease.
