Product Specific Information

The eBio1D4B monoclonal antibody reacts with mouse CD107a (LAMP-1), a type I lysosomal membrane protein that is extensively glycosylated and expressed in late endosomes-lysosomes of all cells. CD107a is transiently expressed on the surface of degranulating cytolytic T cells and is involved in various cellular functions including cancer metastasis and lysosomal storage disorders.

Applications Reported:
Suitable for intracellular staining followed by flow cytometric analysis, immunoprecipitation, western blotting, and immunohistochemical staining (frozen and paraffin-embedded tissues). Fluorochrome-conjugated forms are recommended for intracellular flow cytometry.

Applications Tested:
Validated by intracellular staining and flow cytometric analysis of mouse thioglycolate-elicited peritoneal exudate cells. Recommended use ≤0.5 µg/test (in 100 µL final volume, 10⁵–10⁸ cells/test). Titrate antibody for optimal performance.

Target Information

LAMP1 (CD107a) is a major lysosomal membrane component, with 1–2% present on the plasma membrane. It contains a signal peptide, 18 N-linked glycosylation sites, a single transmembrane domain, and a short cytosolic tail. LAMP1 plays a role in lysosome biogenesis and fusion with phagosomes. Upon activation, LAMP1 translocates to the plasma membrane via a tyrosine-based motif (YXXI). Alteration of this motif disrupts lysosomal localization. LAMP1 and LAMP2 mediate PBMC adhesion to vascular endothelium, suggesting roles in inflammation. Increased LAMP1 expression is observed in neurons and glial cells near senile plaques in Alzheimer’s disease and in activated mast cells.
