Product Specific Information

The TEK4 monoclonal antibody reacts with mouse Tie-2 (CD202), a member of the tyrosine kinase receptor family. Tie-2 is expressed on endothelial and a subset of hematopoietic cells and plays a role in angiogenesis and hematopoiesis during mouse embryonic development. In fetal liver and adult bone marrow, Tie-2 is expressed by hematopoietic progenitor cells (Lineage^-, c-Kit^+, Sca1^+). Long-term multilineage repopulating cells are found in Tie-2^+ populations but not in Tie-2^- cells.

Applications Reported: Suitable for flow cytometric analysis and immunohistology staining of frozen tissue sections.
Applications Tested: Blocking of staining with fluorochrome-conjugated TEK4 on mouse bone marrow cells. Recommended ≤1 µg/test (100 µL final volume). Cell number: 10^5–10^8 cells/test. Antibody should be titrated for optimal performance.

Target Information

The TEK receptor tyrosine kinase is expressed predominantly in endothelial cells. TEK signaling is essential for endothelial-smooth muscle cell communication during venous morphogenesis, and mutations are linked to inherited venous malformations. Angiopoietin-1 binds TEK to mediate vascular development signaling. TIE2 expression increases with thyroid-stimulating hormone and cAMP-elevating agents. Hematopoietic stem cells (HSCs) expressing TIE2 are quiescent, antiapoptotic, and adhere to osteoblasts in the bone marrow niche.
