Product Specific Information

The monoclonal antibody MH9D1 recognizes interleukin-9 (IL-9), a proinflammatory cytokine involved in type 2 immune responses. Recent studies indicate IL-9 is secreted by a distinct T-helper lineage called Th9, derived from Th2 cells with TGF beta or directly from naive CD4+ T cells with TGF beta and IL-4.

IL-9 belongs to the cytokine receptor gamma chain-dependent family (IL-2, IL-4, IL-7, IL-15, IL-21). It affects Th2 lymphocytes, B lymphocytes, mast cells, eosinophils, and epithelial cells, implicating IL-9 in asthma and allergy-related disorders.
Staining with MH9D1 is not blocked by MH9A4, indicating distinct epitopes.

Applications Reported: Intracellular staining and flow cytometric analysis.
Applications Tested: Pre-titrated and tested with Th9 polarized human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00) and BestProtocols® Protocol A.

A test is defined as the amount (µg) of antibody that stains a sample in 100 µL final volume. Cell numbers may range from 10⁵ to 10⁸ cells/test.

Fluorochrome Information:
eFluor® 450 is an alternative to Pacific Blue®, excited at 405 nm and emits at 445 nm. Compatible with Violet laser detection systems.
Filtered post-manufacture with 0.2 µm membrane.


Target Information

Interleukin 9 (IL-9) is produced by activated Th2 lymphocytes and regulates cell types involved in asthma responses, including B/T lymphocytes, mast cells, eosinophils, and epithelial cells.
Mouse IL-9 cDNA encodes a 144 amino acid precursor with an 18 amino acid signal peptide, forming a mature cysteine-rich protein (~14 kDa). Native mouse IL-9 is heavily glycosylated.

TGF beta influences differentiation of IL-17-producing T helper and regulatory T cells by reprogramming Th2 cells to secrete IL-9, driving Th9 cell formation.
