Product Specific Information

The 4B12 monoclonal antibody reacts with mouse CCR7 (CD197), a chemokine receptor for CCL19 and CCL21. CCR7 mediates lymphocyte migration to secondary lymphoid organs and plays a role in dendritic cell and B cell localization during immune responses.

Human CCR7 is a marker for central memory T cells (TCM), involved in thymocyte development. Deficiency causes disturbed thymic architecture and abnormal T cell development.

For optimal CCR7 visualization, multi-color staining and incubation at 37°C are recommended for brighter signals and better resolution.

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse splenocytes and thymocytes, ≤1 µg/test

  1. Prepare cell suspension and block Fc receptors with 5 µg/million cells anti-mouse CD16/32 for 15 min on ice.
  2. Add 1 µg/million cells 4B12 and incubate at 37°C (preferred) or 2–8°C for 30 min.
  3. Wash cells once with Flow Cytometry Staining Buffer and decant supernatant.
  4. Analyze on flow cytometer or proceed with secondary staining.

Note: Co-staining with CCL19-Fc may be hindered due to steric interference. 4B12 binding can block CCL19-Fc binding, but not vice versa.

Target Information

CCR7 is a G protein-coupled chemokine receptor induced by Epstein-Barr virus (EBV). It is expressed in blood, bone marrow, lymph node, and intestine, especially in activated B and T lymphocytes. CCR7 regulates migration of memory T cells and dendritic cell maturation. CCL19 (ECL) is a specific ligand. Receptors of the C-C chemokine family include CCR1–CCR5 and Duffy antigen, critical for T cell chemotaxis and phagocyte migration.
