Product Specific Information

  • Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
  • Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

IL-1 alpha (Interleukin-1 alpha) is a proinflammatory cytokine expressed by monocytes, macrophages, and dendritic cells. It is encoded by the IL1A gene and signals through two receptors, IL-1RI and IL-1RII, shared with IL-1 beta.
Located on chromosome 2q13, IL-1 alpha plays a key role in innate host defense by triggering production of other proinflammatory cytokines and initiating acute-phase responses.
Its activity is regulated by IL-1 Receptor Antagonist (IL-1RA), which blocks receptor binding via competitive inhibition.
IL-1 alpha is involved in immune responses, inflammation, and hematopoiesis. It is synthesized as a proprotein by monocytes and macrophages, processed upon cell injury, and induces apoptosis.
The IL1A gene is part of a cytokine gene cluster on chromosome 2, regulating NF-kappaB and MAPK signaling pathways. IL-1 alpha also stimulates IL6 and COX-2 (PTGS2) expression and enhances nitric oxide (NO) production.
Elevated IL-1 alpha levels are associated with chronic inflammatory diseases such as rheumatoid arthritis, psoriasis, and multiple sclerosis.
