Product Specific Information
The Connexin43 monoclonal antibody recognizes mouse Connexin 43, a gap junction protein facilitating direct cell-cell communication. Gap junctions consist of two hemichannels formed by six connexin proteins. Connexin 43 is expressed in various tissues including the central nervous system, heart, and bone. It is the most abundant connexin in myocardium and primary connexin in astrocytes. Mutations in Connexin 43 are linked to oculodentodigital dysplasia, a developmental disorder with craniofacial and limb abnormalities. Connexin 43 may also serve as a therapeutic target for malignant melanomas and mammary gland tumors.
This monoclonal antibody recognizes human and rat Connexin 43 as well.
Applications Reported: Western blotting, immunohistochemistry (frozen tissue), microscopy, and immunocytochemistry.
Applications Tested: Tested by IHC on frozen mouse tissue at ≤5 µg/mL. Antibody should be titrated for optimal performance.
Purity: >90% (SDS-PAGE).
Aggregation: <10% (HPLC).
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
Connexin 43 (Cx43) is a gap junction protein forming hexameric hemichannels for intercellular communication. Gap junctions regulate development and cell growth. Phosphorylation of Cx43 by PKC (Ser368) and Src modulates its function. Connexin 43 is crucial for synchronized heart contraction and embryonic development. A related pseudogene, GJA1P, is located on chromosome 5. Mutations in GFAP gene are associated with Charcot-Marie-Tooth disease, oculodentodigital dysplasia, and heart malformations. Alternatively spliced GFAP transcript variants have been identified.
