The NRRF-30 antibody reacts with mouse Foxp3 (FORKHEAD BOX P3, SCURFIN, JM2). Cross-reactivity with other proteins has not been determined. Foxp3 is a 49–55 kDa transcriptional regulator, identified as the gene defective in scurfy (sf) mice. High expression of Foxp3 mRNA is found in CD4+/CD25+ regulatory T cells (Treg), and ectopic expression in CD4+/CD25− cells induces a Treg phenotype.

Immunoblotting with NRRF-30 maps the epitope to amino acids 1–75 of mouse Foxp3. Intracellular staining of mouse splenocytes using fluorochrome-conjugated NRRF-30 with eBioscience Foxp3 Staining Buffers reveals approximately 3% of total cells in C57Bl/6 and 5% in BALB/c strains. Multicolor flow cytometry shows ~90% of CD4+/CD25+ cells and 4% of CD4+/CD25− cells stained with NRRF-30. Co-staining with FJK-16s (anti-mouse/rat Foxp3, Product #71-5775-40) shows 100% correlation.

Target Information

FOXP3 (Forkhead box protein 3) is a transcriptional regulator essential for immune homeostasis. It is 381 amino acids long and highly expressed in CD25+CD4+ regulatory T cells, weakly in CD4+CD25− cells, and absent in CD4−CD8+ T cells. FOXP3 acts as a master regulatory gene and marker of regulatory T cells. Defects cause the scurfy phenotype in mice and IPEX syndrome in humans (immune dysfunction, polyendocrinopathy, enteropathy, X-linked). FOXP3 expression in human cancer cells is regulated by p53 in response to DNA damage.
