This TXRX10 monoclonal antibody reacts with human and mouse TOX (Thymocyte selection-associated HMG box protein).
TOX is a DNA-binding nuclear factor expressed in thymocytes, T lymphocytes, NK cells, and lymphoid tissue-inducer (LTi) cells.
Its expression is upregulated during beta-selection and positive selection in thymocytes.

Studies using Tox knockout mice show that TOX plays a critical role in CD4⁺ T cell development, regulating the CD4^loCD8^lo to CD4⁺CD8^lo transition.
TOX is also involved in NK and LTi cell development and is induced by calcineurin-mediated TCR signaling during positive selection.

Applications Reported: Immunoprecipitation and Western blotting (WB).
Applications Tested: Tested by immunoblotting of reduced mouse thymocyte lysate ≤ 5 µg/mL.
Careful titration recommended for optimal assay performance.

Target Information

TOX (Thymocyte selection-associated high mobility group box protein) is a member of the HMG box family of DNA-binding proteins.
It regulates T-cell development and is a 526 amino acid nuclear protein.
TOX expression is upregulated by pre-TCR and TCR activation in immature thymocytes.
TOX-mediated positive selection is associated with RUNX3 up-regulation and is calcineurin dependent.
TOX-dependent transition to the CD4⁺CD8 stage is essential for development of class II MHC-specific T cells.
