Product Specific Information

This antibody was cross-adsorbed to a non-specific phospho-tyrosine peptide then affinity-purified using phospho-SHP1 (Tyr-536) peptide.
It detects a 68 kDa band on SDS-PAGE immunoblots of human Jurkat cells treated with pervanadate, but is not observed in control cells.

Target Information

SHP-1 (PTPN6) is a non-receptor protein tyrosine phosphatase primarily expressed in hematopoietic cells.
The enzyme consists of two SH2 domains, a catalytic domain, and a regulatory domain. SHP-1 downregulates several tyrosine kinase-regulated pathways by dephosphorylating target proteins.
In hematopoietic cells, its amino-terminal SH2 domain binds to tyrosine-phosphorylated erythropoietin receptors (EpoR) to negatively regulate hematopoietic growth.
Overexpression in epithelial cells leads to dephosphorylation of the Ros receptor tyrosine kinase, reducing Ros-dependent cell proliferation and transformation.
In myeloid cells, SHP-1 associates with the IL-3R beta chain following ligand binding and downregulates IL-3-induced tyrosine phosphorylation and proliferation.
