Product Specific Information

This NGZB monoclonal antibody reacts with mouse Granzyme B, a serine protease found in cytotoxic T cells and NK cells. Granzyme B (CGL1, CTLA-1) is essential for inducing apoptosis in target cells through mannose 6-phosphate receptor-mediated pathways.
This antibody does not cross-react with human Granzyme B and recognizes a distinct epitope.

Applications Reported: Intracellular staining and flow cytometric analysis.
Applications Tested: Tested by intracellular staining followed by flow cytometric analysis of stimulated mouse splenocytes using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) and BestProtocols® Protocol A.
Recommended use ≤ 0.125 µg per test (100 µL final volume, 10⁵–10⁸ cells/test). Titrate for optimal performance.

Fluorophore Information:
eFluor® 450 is an alternative to Pacific Blue®. Excitation 405 nm; Emission 445 nm; Laser: Violet.
Post-manufacturing filtered (0.2 µm).

Target Information

Granzyme B is a member of the granzyme serine protease family located in cytotoxic T cells and NK cells. It induces apoptosis through caspase-3 activation and mitochondrial pathways (Bid cleavage).
Granzymes (A–G) have been identified in mouse CTLs and NK cells; fewer are known in humans.
