This CTSREVL monoclonal antibody recognizes human GPNMB (also known as Osteoactivin, DC-HIL, HGFIN).
GPNMB is a type I transmembrane glycoprotein involved in bone development, tumor progression, neurobiology, and immune regulation. It is expressed on macrophages and dendritic cells and acts as a ligand for Syndecan-4 (SD-4) on T cells, inhibiting their activation and proliferation. Expression of GPNMB may indicate tolerogenic myeloid cell properties.

GPNMB expression is observed in various cancers and may contribute to immune evasion. IL-10 and immunosuppressive drugs such as dexamethasone upregulate its expression on macrophages and dendritic cells. The protein may also be secreted.

Target Information

GPNMB encodes a type I transmembrane glycoprotein homologous to pMEL17, a melanocyte-specific protein. It is expressed in low-metastatic melanoma cell lines and xenografts but absent in highly metastatic ones.
GPNMB may play a role in growth delay and reduced metastatic potential. Two transcript variants encoding different isoforms have been identified.
