Product Specific Information

Description:
The MP6-XT22 antibody reacts with mouse tumor necrosis factor-alpha (TNF alpha), a 17 kDa cytokine produced by monocytes, macrophages, neutrophils, NK cells, and CD4(+) T cells. TNF alpha exhibits cytolytic activity against various tumor cells and plays a key role in immune regulation. It exists in soluble (17 kDa) and membrane-bound (26 kDa) forms.

Applications Reported:

• Intracellular staining followed by flow cytometric analysis.
• Internal testing shows Alexa Fluor 561 may non-specifically stain B cells in Swiss Webster and SJL mice, but not in BALB/c or C57BL/6 strains.

Applications Tested:

• Tested by intracellular staining and flow cytometric analysis of stimulated mouse splenocytes using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00).
• Recommended use ≤ 0.25 µg per test (100 µL final volume).
• Cell number: 10⁵–10⁸ cells/test (empirical optimization recommended).
• Carefully titrate for optimal performance.

• Alexa Fluor 561 emits at 575 nm.
• Suitable for spectral cytometers; compatible with PE and PE-eFluor 610.

Target Information

TNF alpha is a multifunctional proinflammatory cytokine in the TNF superfamily, primarily secreted by macrophages. It binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR and regulates immune cell activity, proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. TNF alpha forms multimers (dimers, trimers, pentamers) and appears as a 17 kDa band under non-reducing SDS-PAGE conditions. It has roles in immune defense against infections and tumor necrosis. TNF alpha is implicated in autoimmune diseases, insulin resistance, and cancer.
