Product Specific Information

The monoclonal antibody A1exF5 recognizes both human and mouse Arginase 1 (Arg1), a cytosolic enzyme. This clone is compatible with standard intracellular staining protocols and the Foxp3/Transcription Factor Staining Buffer Set.

Applications Reported:

• Intracellular staining followed by flow cytometric analysis.

Applications Tested:
Tested by intracellular staining of normal human lysed whole blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00).
Recommended use ≤ 1.0 µg/test (100 µL final volume).
Cell number: 10⁵–10⁸ cells/test.
Antibody titration is recommended for optimal results.

Fluorochrome Information:
Brilliant Violet™ 421 (BV421) emits at 423 nm and is excited by the 405 nm violet laser. Ensure the cytometer is equipped to detect this fluorochrome.

Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product #SB-4401-42) or Brilliant Stain Buffer™ (Product #00-4409-75) to minimize non-specific interactions.

Target Information

Arginase-1 (Arg1) is a 35 kDa enzyme that converts L-arginine to urea and L-ornithine, the final step in the urea cycle. The resulting polyamines are essential for cell proliferation and detoxification.
By degrading arginine, Arginase 1 down-regulates nitric oxide production.
Arginase 1 is expressed in liver, neutrophils, myeloid-derived suppressor cells (MDSC), and neural stem cells.
In mice, it is a hallmark of alternatively activated macrophages (M2a).
Arginase 1 is often expressed in tumor-infiltrating myeloid cells and hepatocellular carcinomas.
Defects in Arginase 1 cause argininemia, an autosomal recessive disorder characterized by hyperammonemia.
