Target Information
BACE1 (beta-secretase)는 Alzheimer’s disease(AD)에서 아밀로이드 베타 펩타이드(Abeta) 생성을 담당하는 주요 효소입니다. 일부 조기 발병형 AD는 APP(amyloid precursor protein)의 ‘Swedish’ 돌연변이로 인해 BACE1에 의한 단백질 절단이 증가하여 발생합니다. 이러한 병리학적 근거로 인해 BACE1 및 다른 APP 절단 효소(예: gamma-secretase)에 대한 억제제가 치료적 접근법으로 연구되고 있습니다. APP는 먼저 β-secretase에 의해 절단되어 용해성 단백질 파생물과 막 결합형 C99 단편을 생성하며, C99는 γ-secretase의 기질로 작용하여 4 kDa 크기의 amyloid-β 펩타이드를 생성합니다. BACE1은 이러한 β-secretase로 확인된 새로운 막관통성 아스파르트산 프로테아제입니다.
