This antibody is supplied in a ready-to-use formulation.
Recommended positive control tissue: Tonsil (other tissues may also be used).

The antibody is intended for professional laboratory use for detection of Factor XIIIa in formalin-fixed, paraffin-embedded tissue sections using manual qualitative IHC. It recognizes an 83 kDa protein, identified as Factor XIIIa, found in platelets, megakaryocytes, fibroblast-like mesenchymal or histiocytic cells, monocytes, macrophages, and dermal dendritic cells.

Diagnostic Utility:

  • Positive in dermatofibroma (almost all cases)
  • Variable in dermatofibrosarcoma protuberans and desmoplastic malignant melanoma
  • Also positive in capillary hemangioblastoma, hemangioendothelioma, hemangiopericytoma, xanthogranuloma, xanthoma, hepatocellular carcinoma, glomus tumor, and meningioma

Recommended Procedure:

  • Use with formalin-fixed, paraffin-embedded sections
  • Perform heat-induced or enzymatic epitope retrieval before staining
  • IHC staining involves sequential application of primary antibody, secondary antibody, enzyme complex, and chromogenic substrate, with washing steps between each stage
  • Visualization by light microscopy assists in differential diagnosis of pathological processes

Quality Control:

  • Run a positive tissue control with each staining procedure
  • Positive control tissue should contain both positive and negative staining components
  • External positive control specimens should be freshly fixed and processed similarly to patient samples
  • Weakly positive tissues are recommended for detecting subtle changes in antibody sensitivity
  • Internal or external negative controls may be used to verify absence of non-specific staining

Target Information

The Factor XIII A subunit gene encodes a component of the final zymogen in the coagulation cascade. Plasma Factor XIII is a heterotetramer (2 A subunits + 2 B subunits). The A subunits provide catalytic function, while the B subunits act as carrier molecules. Platelet Factor XIII consists only of A subunits.

Upon activation by thrombin and calcium ions, Factor XIIIa catalyzes γ-glutamyl-ε-lysine crosslinking between fibrin molecules, stabilizing the fibrin clot. It also crosslinks α2-plasmin inhibitor and fibronectin to fibrin.
Deficiency in Factor XIII (Type I or Type II) leads to bleeding tendency, poor wound healing, and habitual abortion.
