Recommended positive control tissue: Prostate (not limited to this tissue type).
The primary antibody is intended for professional laboratory use in detecting NKX3.1 protein in formalin-fixed, paraffin-embedded tissue via manual qualitative IHC. It is used after tumor diagnosis by conventional histopathology.
NKX3.1 is a prostate-specific transcription factor involved in normal prostate development and carcinogenesis. It acts as a tumor suppressor gene located on chromosome 8p21.2, often showing loss of heterozygosity in prostate cancer. Its expression is highly restricted to prostate epithelial cells, making it a valuable biomarker for identifying prostate cancer and metastatic lesions of prostatic origin. NKX3.1 shows greater sensitivity than PSA for detecting metastatic prostatic adenocarcinoma, supporting its use with other prostate-restricted markers for differential diagnosis in poorly differentiated carcinomas.
For optimal staining:
- Use formalin-fixed, paraffin-embedded sections.
- Apply heat-induced or enzymatic epitope retrieval before staining.
- IHC staining involves sequential application of primary antibody, secondary antibody, enzyme complex, and chromogenic substrate, with washing steps.
- The visible reaction product at the antigen site is interpreted under a light microscope for diagnostic evaluation.
Positive Control:
Run a positive tissue control with each staining procedure. Tissues should include both positive and negative staining cells. External positive control materials should be freshly prepared and processed similarly to patient samples. Use weakly positive tissues to detect subtle changes in antibody sensitivity and reagent stability.
Negative Control:
Internal or external negative control tissues may be used. Non-staining components should confirm absence of specific staining and indicate background levels. If specific staining appears in negative controls, results must be considered invalid.
Target Information
NKX3-1 is a homeodomain-containing transcription factor and a putative prostate tumor suppressor. It is expressed in an androgen-regulated, prostate-specific manner. Loss of NKX3-1 expression is commonly observed in human prostate carcinoma and prostatic intraepithelial neoplasia.
