Target Information

Cluster of differentiation 8 (CD8), a type I transmembrane glycoprotein of the immunoglobulin family, plays an integral role in signal transduction, T cell differentiation, and activation.
CD8 is predominantly expressed on T cells as a disulfide-linked heterodimer of CD8α and CD8β, functioning as a co-receptor with the T cell receptor (TCR) for MHC class I molecules.
Its counterpart, CD4, acts as a co-receptor for MHC class II molecules. CD8α is essential for MHC-I binding and is also expressed as a homodimer on subsets of T cells, NK cells, monocytes, and dendritic cells.
Engagement of MHC-I/peptide complexes triggers recruitment of Lck kinase, leading to lymphokine production, motility, and activation of cytotoxic T lymphocytes (CTLs).
Activated CTLs are key in pathogen and tumor cell clearance. Differentiation of naïve CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
