MA3-913은 인간, 마우스, 랫, 개, 돼지, 닭, 토끼의 골격근 조직에서 calsequestrin을 검출합니다. 이 항체는 type I(느린) 및 type II(빠른) 골격근 섬유 모두에서 calsequestrin을 인식합니다.
Western blot에서 63 kDa의 단백질 밴드를 검출하며, 이는 개 골격근 추출물의 calsequestrin을 나타냅니다. 또한 더 높은 분자량의 밴드는 sarcoplasmic reticulum 내의 calsequestrin 유사 단백질로 추정됩니다.
형광 염색 패턴은 Z-line 양쪽의 triad 쌍에 해당하는 이중 행의 형광 점으로 나타납니다.
항원은 정제된 토끼 골격근 sarcoplasmic reticulum입니다.
Target Information
Sarcoplasmic reticulum(SR)은 심장 및 골격근에서 세포 내 칼슘 수준을 조절하기 위해 칼슘을 저장하고 방출하는 역할을 합니다. SR 내강에는 여러 칼슘 결합 단백질이 존재하며, 그중 가장 대표적인 것이 calsequestrin입니다.
Calsequestrin은 근육 세포의 이완 중 SR 내에 축적된 칼슘을 결합하여 저장하는 단백질로, terminal cisternae의 접합면 근처에 칼슘을 국소화시키는 기능을 수행합니다.
이 단백질은 고도로 산성이며, 높은 결합 용량과 중간~낮은 친화도를 가진 칼슘 결합 특성을 가집니다.
