Product Specific Information
The L243 monoclonal antibody reacts with human MHC class II molecule HLA-DR, expressed on antigen-presenting cells (APC) such as B cells, monocytes, macrophages, dendritic cells, and activated T cells.
HLA-DR is a heterodimeric transmembrane protein composed of alpha and beta subunits, crucial for peptide presentation to CD4⁺ T lymphocytes.
L243 recognizes a distinct epitope from LN3 and does not cross-block its binding.
Applications Tested:
Pre-titrated for flow cytometric analysis of normal human peripheral blood cells. Recommended use: 5 µL (0.06 µg)/test in 100 µL final volume. Cell count may range from 10⁵–10⁸ cells/test.
Fluorochrome Information:
PerCP-eFluor® 710 replaces PE-Cy5 or PerCP-Cy5.5, emits at 710 nm, excited by 488 nm blue laser.
Recommended filters: 685 LP dichroic mirror and 710/40 bandpass filter (695/40 acceptable alternative).
Stable after fixation with 2% formaldehyde overnight at 4°C.
Excitation: 488 nm; Emission: 710 nm; Laser: Blue Laser.
Post-manufacturing filtration: 0.2 µm.
Target Information
HLA-DR is a transmembrane glycoprotein composed of a 36 kDa alpha chain (DRA) and 27 kDa beta chain (DRB).
The alpha chain gene (DRA) has 5 exons encoding leader peptide, extracellular domains, transmembrane domain, and cytoplasmic tail.
DRA acts as the sole alpha chain for DRB1, DRB3, DRB4, and DRB5, while DRB1 alleles define peptide-binding specificity.
HLA-DR is expressed on APCs such as B lymphocytes, monocytes, macrophages, thymic epithelial cells, and activated T lymphocytes.
MHC class II molecules (DR, DQ, DP) present processed peptides to T-helper cells, initiating immune responses.
HLA and MHC antibodies are essential in Immunopeptidomics for neoantigen identification via LC-MS/MS.
