Product Specific Information

The 122 monoclonal antibody reacts with mouse B7-DC (PD-L2), a member of the B7 family within the immunoglobulin superfamily. B7-DC has a predicted molecular weight of approximately 25 kDa and a short cytoplasmic tail (4 amino acids). It is primarily expressed in dendritic cells and monocytes/macrophages.

Unlike other B7 family members, B7-DC does not bind CD28/CTLA-4 but serves as a ligand for PD-1. The PD-1 and B7-DC/PD-L2 interaction can either costimulate or suppress T cell proliferation depending on activation state. Clone 122 blocks binding of TY25, another mAb specific for mouse B7-DC.

Applications Reported: Flow cytometric analysis.
Applications Tested: Mouse bone marrow-derived dendritic cells; ≤ 0.5 µg/test (100 µL final volume, 10⁵–10⁸ cells/test).
Careful titration is recommended for optimal performance.

Fluorophore Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with the blue laser (488 nm). Recommended filter: 710/50 bandpass (695/40 acceptable alternative). Ensure instrument compatibility.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-822-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark. Clone-specific performance should be empirically determined.

Target Information

Programmed death-ligand 2 (PD-L2, B7-DC) is a B7 ligand family member acting as a ligand for PD-1. Expressed primarily in dendritic cells, its expression can be induced on various immune and non-immune cells under specific cytokine environments (IL-4, TNF-α, IFN-γ).

PD-L2 has higher affinity to PD-1 than PD-L1 and functions as an inhibitory immune checkpoint molecule, regulating T cell activation and cytokine secretion. PD-L2 can also stimulate T cell proliferation independently of PD-1, interacting with a second receptor, RGMb, involved in respiratory tolerance regulation.

Human PD-L2 monomer consists of:

• 201-aa extracellular domain
• 21-aa transmembrane domain
• 32-aa cytoplasmic domain
