Tumor necrosis factor (TNF) receptor associated factors (TRAFs) are adaptor proteins that link the TNF receptor superfamily to signaling pathways, regulating cell death and stress responses.
TRAF proteins share a homology region enabling them to bind to cell receptors and other TRAF proteins, activating different signaling cascades.

For example:

• TRAF2 and TRAF3 bind to CD40, a receptor involved in B cell immunity, critical for NF-κB activation in mouse B lymphocytes.
• TRAF2 and TRAF6 are required for CD40 signaling in non-hematopoietic cells.
• TRAF2 interacts with lymphotoxin-beta receptor (LTβR) with TRAF3, cIAP1, and Smac, influencing apoptosis regulation.
