Product Specific Information

The OKT3 monoclonal antibody reacts with an epitope on the epsilon-subunit within the human CD3 complex. It exhibits potent immunosuppressive properties and is effective in the treatment of renal, heart, and liver allograft rejection. CD3 subunits (gamma, delta, epsilon) are essential for TCR complex assembly and surface expression. CD3 is expressed by thymocytes and all mature T cells.

Crosslinking of TCR initiates intracellular signaling leading to activation and proliferation.
This antibody is validated for flow cytometric analysis of normal human peripheral blood cells at 5 µL (1 µg) per test (100 µL final volume).
Cell number may vary from 10⁵ to 10⁸ cells per test.

Fluorophore Information:
eFluor® 450 is an alternative to Pacific Blue®, excited at 405 nm and emits at 445 nm. Ensure instrument compatibility with violet laser detection.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

The CD3 complex transduces antigen-recognition signals into T cells and regulates surface expression of the TCR complex. CD3 subunits (gamma, delta, epsilon, zeta) are structurally related members of the immunoglobulin superfamily encoded on chromosome 11. They associate with cytoplasmic signal transduction molecules via tyrosine-based motifs.

CD3 is expressed on 68–82% of peripheral blood lymphocytes and 65–85% of thymocytes, but not on B or NK cells. Decreased T lymphocytes may occur in autoimmune diseases.
Defects in CD3 genes are linked to CD3 immunodeficiency.
