Product Specific Information
This MR9-4 monoclonal antibody reacts with the mouse T cell receptor (TCR) V beta 5.1/5.2 chain. The MR9-4 antibody recognizes the V beta 5.1 and 5.2 chains on T cells from mouse strains with the b haplotype of the Tcrb gene, including C57BL/6, B10, and C58/J. V beta 5.1 and 5.2 are deleted in mouse strains that express MHC Class II I-E, including Balb/c, C3H, and DBA/2. Reports indicate that V beta 5+ T cells recognize the OVA peptide.

Applications Tested: Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤ 0.25 µg per test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best performance.

Target Information
T cell receptors (TCR) discriminate foreign from self-peptides presented by MHC class II molecules, critical for adaptive immunity. Recognition of self-peptides is linked to autoimmune diseases; mutant self-peptides are associated with tumors. TCR engagement by bacterial superantigens causes toxic shock syndrome. Autoantibodies to V beta segments have been isolated from patients with RA and SLE, blocking TH1-mediated inflammation. Most human T cells express TCR alpha-beta with CD4 or CD8 (SP), while a subset lacks both (DN), which increases in autoimmune and immunodeficiency disorders. Gamma-delta T cells, mainly epithelial, show limited diversity and distinct antigen recognition, with known antitumor and immunoregulatory roles.
