Product Specific Information

This antibody was cross-adsorbed to unphosphorylated Asc (Tyr-144) peptide before affinity purification using phospho-Asc (Tyr-144) peptide.
It detects phosphorylated Asc (Tyr-144) in the inflammasome. In J774 macrophage cells primed with LPS and treated with nigericin, the antibody colocalized with an inflammasome marker, caspase-1 inhibitor (FAM-YVAD-FMK).
Additionally, it detects a 20 kDa Asc protein in western blots of J774A.1 mouse macrophages treated with pervanadate.

Target Information

Functions as a key mediator in apoptosis and inflammation. Promotes caspase-mediated apoptosis involving predominantly caspase-8 and caspase-9 in a cell type-specific manner.
Involved in activation of the mitochondrial apoptotic pathway and promotes proteolytic maturation of BID independently of FADD.
Participates in macrophage pyroptosis, a caspase-1-dependent inflammatory form of cell death, and serves as the major constituent of the ASC pyroptosome.
Acts as an adapter in inflammasome assembly, leading to activation of caspase-1 and secretion of proinflammatory cytokines.
Mediates recruitment of caspase-1 to inflammasomes containing receptors such as NLRP2, NLRP3, AIM2, and possibly IFI16.
Also involved in adaptive immunity, dendritic cell maturation, and transcriptional activation of cytokines and chemokines via AP-1, NF-κB, and MAPK signaling pathways.
For regulation of NF-κB, both activating and inhibitory functions have been reported.
