Product Specific Information

The eBioRDR5 monoclonal antibody reacts with human CD127 (Interleukin-7 Receptor alpha). CD127 complexes with CD132 (common gamma chain) to form the IL-7 receptor (IL-7R), a type I glycoprotein (75–80 kDa). It is expressed on immature B cells, thymocytes, and most mature T cells, with transient down-regulation upon activation.

IL-7 binding activates signaling via tyrosine kinase pathways including Jak/STAT, essential for lymphocyte development and T-cell homeostasis. CD127 expression is notably down-regulated on CD4+CD25+ regulatory T cells (Tregs), making it a marker for identifying functional Tregs expressing Foxp3.

Binding of eBioRDR5 to PBMCs is blocked by pre-incubation with recombinant human IL-7 (Product #14-1079-80).

Applications Tested: Pre-diluted, tested on normal human peripheral blood cells at 5 µL (0.25 µg)/test (in 100 µL final volume, 10⁵–10⁸ cells/test).

Alexa Fluor 561 emits at 575 nm and is optimized for use on spectral cytometers, multiplexed with PE and PE-eFluor 610.

Target Information

CD127 (Interleukin-7 Receptor α, IL7Rα) regulates lymphopoiesis. The active receptor is an α/γ chain heterodimer, with the γ chain activating signal transduction and the α chain mediating specific signaling. CD127 promotes proliferation of precursor B cells, thymocytes, and mature T cells.
Dysfunction of CD127 is associated with severe combined immunodeficiency (SCID).
