Product Specific Information

The A7R34 monoclonal antibody reacts with mouse CD127, the high-affinity alpha subunit of the mouse IL-7 receptor.
IL-7 receptor alpha chain is expressed by immature B cells in bone marrow, double-negative (CD4⁻CD8⁻), single-positive (CD4⁺ and CD8⁺), but not double-positive (CD4⁺CD8⁺) thymocytes.
In peripheral tissues, mature T cells express CD127 at low levels. A7R34 inhibits binding of IL-7 to its receptor and has been used in vivo and in vitro to study IL-7’s role in T and B cell development and activation.
Binding of A7R34 blocks the binding of SB/199, another antibody recognizing mouse CD127.

Applications Reported: Flow cytometric analysis.
Applications Tested: Flow cytometric analysis of mouse splenocytes at ≤0.25 µg per test (100 µL final volume, 10⁵–10⁸ cells/test). Antibody should be titrated for optimal performance.

Alexa Fluor 561 emits at 575 nm and is intended for use on spectral cytometers. It can be multiplexed with PE and PE-eFluor 610.

Target Information

CD127 (Interleukin-7 receptor alpha, IL7Rα) is a glycoprotein regulating lymphopoiesis. The active IL7R complex is an alpha/gamma chain heterodimer.
The gamma(c) chain activates signal transduction, while the alpha chain mediates specific signaling events.
CD127 promotes proliferation of precursor B cells, thymocytes, T cell progenitors, and mature CD4⁺/CD8⁺ T cells.
Dysfunction of CD127 is associated with severe combined immunodeficiency (SCID) and related T⁻B⁻NK⁺ SCID phenotypes.
