• Cloning partner: Sp/2
• Reacts with the 160 kD polypeptide subunit of human neurofilament.
• Recognizes a phosphate-independent epitope in the tail (carboxy) domain of NF-M in most vertebrates and invertebrates.
• Suitable for immunohistochemical staining of Bouin’s and alcohol-fixed paraffin-embedded or frozen tissue sections.
• Recommended incubation: 30–60 minutes at room temperature or overnight at 4°C.

Target Information

Neurofilaments (NFs) are intermediate filaments expressed almost exclusively in neuronal cells, particularly in large axons.
They are composed of three polypeptide chains: NF-M (medium), NF-H (high), and NF-L (light).
These share a coiled-coil core domain but differ in N- and C-terminal sequences.
NF-M and NF-H have long, flexible tails that mediate filament interactions and cytoskeletal architecture.
NF-M phosphorylation and O-GlcNAcylation are reciprocally regulated, affecting translocation and filament formation.
Antibodies to NF subunits are valuable neuronal markers due to their abundance, neuron-specific expression, and biochemical stability.
