Target Information
Glycogen synthase kinase-3 (GSK3)는 원래 glycogen synthase의 조절 인자로 확인된 단백질 키나아제입니다. 이후 단백질 합성, 세포 증식, 분화, 미세소관 조립/해체, 세포사멸 등 다양한 세포 기능의 조절에 관여함이 밝혀졌습니다.
GSK3는 고유한 기질 특이성을 가지며, 인산화는 GSK 인산화 부위의 C-말단에 phosphoserine 또는 phosphotyrosine이 존재할 때만 일어납니다.
GSK3에는 α와 β 두 가지 isoform이 존재하며, 이들은 인산화에 의해 엄격히 조절됩니다.
- GSK3β의 Ser9 (또는 GSK3α의 Ser21) 인산화는 PKB에 의해 발생하며, 키나아제의 비활성화에 관여합니다.
- GSK3β의 Tyr216 (또는 GSK3α의 Tyr279) 인산화는 활성화에 필수적입니다.
활성화된 GSK3β는 p53, c-Myc, c-Jun, HSF-1, cyclin D1 등 다양한 단백질을 인산화하며, tau 단백질의 비정상적 인산화에도 관여하여 알츠하이머병 진행과 관련이 있습니다.
