Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Rat IL-10. Anti-Rat IL-10-specific antibody was purified by affinity chromatography and then biotinylated.

Sandwich ELISA:
To detect Rat IL-10 by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.25–1.0 µg/mL of this antibody is required. This biotinylated polyclonal antibody, in conjunction with PeproTech Polyclonal Anti-Rat IL-10 (500-P139) as a capture antibody, allows detection of at least 0.2–0.4 ng/well of Recombinant Rat IL-10.

Western Blot:
To detect Rat IL-10 by Western Blot analysis, use at 0.1–0.2 µg/mL. In conjunction with compatible secondary reagents, the detection limit for Recombinant Rat IL-10 is 1.5–3.0 ng/lane under reducing or non-reducing conditions.

Target Information

IL-10 encodes a cytokine primarily produced by monocytes and some lymphocytes. It regulates immune responses and inflammation by down-regulating Th1 cytokines, MHC class II antigens, and costimulatory molecules on macrophages. It enhances B cell survival, proliferation, and antibody production, blocks NF-kappa B activity, and regulates the JAK-STAT signaling pathway. Mouse knockout studies indicate IL-10 is essential for intestinal immune regulation. Mutations are linked to susceptibility to HIV-1 infection and rheumatoid arthritis. Diseases associated with IL-10 include Graft-Versus-Host Disease and HIV-1.
