Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Rat IL-10. Anti-Rat IL-10-specific antibody was purified by affinity chromatography employing an immobilized Rat IL-10 matrix.

Sandwich ELISA:
To detect Rat IL-10 by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.5–2.0 µg/mL of this antibody is required. This antigen affinity purified antibody, in conjunction with PeproTech Biotinylated Anti-Rat IL-10 (500-P139Bt) as a detection antibody, allows the detection of at least 0.2–0.4 ng/well of Recombinant Rat IL-10.

Western Blot:
To detect Rat IL-10 by Western Blot analysis, this antibody can be used at a concentration of 0.1–0.2 µg/mL. Used with compatible secondary reagents, the detection limit for Recombinant Rat IL-10 is 1.5–3.0 ng/lane, under either reducing or non-reducing conditions.

Target Information

IL-10 encodes a cytokine primarily produced by monocytes, with additional production by lymphocytes. This cytokine regulates immune response and inflammation by down-regulating Th1 cytokines, MHC class II antigens, and costimulatory molecules on macrophages. It enhances B cell survival, proliferation, and antibody production, and can block NF-kappa B activity. IL-10 is involved in the JAK-STAT signaling pathway regulation. Knockout studies in mice indicate its essential role in intestinal immune regulation. Mutations in IL-10 are associated with susceptibility to HIV-1 infection and rheumatoid arthritis. Related diseases include Graft-Versus-Host Disease and HIV Type 1.
