Product Specific Information
PA1-1961 detects PA200 from bovine, human, mouse, and rat samples.
It has been successfully used in Western blot, immunofluorescence, and immunoprecipitation assays.
- Western blot: Detects ~200 kDa PA200 protein in bovine testis extract. In mouse tissues, detects both 200 kDa (testis-enriched) and 160 kDa (spleen-enriched) proteins.
- Immunofluorescence: In HeLa cells, PA200 shows diffuse nuclear staining before ionizing radiation, and focal nuclear aggregates after treatment.
- Immunogen: Synthetic peptide corresponding to residues T(1620)–G(1634) of human PA200.
Target Information
PA200 (PSME4) is an associated proteasome component that recognizes acetylated histones and promotes ATP- and ubiquitin-independent degradation of core histones during spermatogenesis and DNA damage response.
It binds acetylated histones via its bromodomain-like region and activates the proteasome by opening the gated channel for substrate entry.
As part of the spermatoproteasome, PA200 promotes histone degradation and exchange during spermatogenesis and contributes to DNA damage response by facilitating histone degradation following double-strand breaks.
