Target Information
EIF2AK2 (PKR) is one of four kinases that specifically phosphorylate Ser51 of translation initiation factor eIF2-alpha in response to various environmental stresses.
PKR plays a key role in antiviral defense through its activation by double-stranded RNA produced during viral infection.
It is involved in IFN-induced innate antiviral response, virus-induced apoptosis, and regulation of cell growth and differentiation.
PKR mediates apoptosis via Fas expression and the FADD/Caspase-8 signaling pathway, and regulates multiple pathways including NF-kappaB activation, p53, p38, and PDGF signaling.
It has been implicated in tumor suppression, malignancy, and several neurodegenerative diseases such as Alzheimer’s, Huntington’s, and amyotrophic lateral sclerosis (ALS).
Viruses have evolved mechanisms to inhibit PKR activity, such as the influenza virus NS1 protein.
