Target Information

Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, when bound to estrogen, undergoes a conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA. This interaction regulates gene transcription through coactivator or corepressor recruitment.

There are two main forms of estrogen receptors, alpha (ESR1) and beta (ESR2), encoded by separate genes. Due to alternative RNA splicing, at least four estrogen receptor-alpha isoforms exist (66 kDa, 53 kDa, 47 kDa, 35 kDa). Estrogen receptors are widely expressed and are essential for sexual development, reproductive function, and bone maintenance. They are also implicated in diseases such as breast cancer, endometrial cancer, and osteoporosis.
