Product Specific Information

This HP-3G10 monoclonal antibody reacts with CD161 (NKR-P1A), a member of the C-type lectin superfamily. CD161 is expressed on natural killer (NK) cells and various T cell subsets, including memory/effector CD4⁺, CD8⁺, γδTCR T cells, and a subset of CD3⁺ thymocytes. It is also detected on human Th17 CD4⁺ T cells. The receptor’s function is not fully understood but may have a stimulatory role. Lectin-like transcript-1 (LLT1) has been identified as its ligand.

Target Information

KLRB1 (CD161) is a type II membrane protein expressed on NK cells and a subset of CD3⁺ T cells. It forms a homodimeric structure (40–44 kDa chains) and plays an inhibitory role in NK cell function. Activation of KLRB1 stimulates acid sphingomyelinase/SMPD1, increases intracellular ceramide, and activates AKT1/PKB and RPS6KA1/RSK1 pathways, promoting T cell proliferation. KLRB1 binds to CLEC2D/LLT1 and specific carbohydrate epitopes, modulating NK cell cytotoxicity and interferon-gamma secretion.
