Product Specific Information
The monoclonal antibody CXCR3-173 recognizes mouse CD183 (CXCR3), a seven-transmembrane G-protein-linked chemokine receptor binding CXCL9 (Mig), CXCL10 (IP-10), and CXCL11 (ITAC). CD183 plays a role in CD4 T cell responses to grafts, and CXCR3 knockout mice show compromised allograft rejection. Expression is found on NK cells, subsets of T lymphocytes, Tregs, and Th1-polarized cells.
The antibody CXCR3-173 affects chemotaxis in response to ligand; ligand presence eliminates staining. In vivo addition delays cardiac and pancreatic islet allograft rejection.

Target Information
This gene encodes a G protein-coupled receptor selective for three chemokines: IP10, Mig, and I-TAC. These belong to the CXC chemokine subfamily, characterized by conserved cysteine residues. Binding induces leukocyte traffic responses, including integrin activation, cytoskeletal changes, and chemotactic migration. Inhibition by Bordetella pertussis toxin suggests coupling with Gi-class G proteins. Signal transduction likely involves enzymes common to other chemokine receptors.
Chemokine-induced desensitization occurs via phosphorylation-dependent receptor internalization, followed by recycling after dephosphorylation. Expression is prominent in effector/memory T cells and inflamed tissues. CXCR3 and its ligands are implicated in inflammatory cell recruitment, making it a potential target for small molecule antagonists to treat inflammatory diseases. Multiple transcript variants exist.
