Product Specific Information
The eBioRDR5 monoclonal antibody reacts with human CD127 (Interleukin-7 Receptor alpha). CD127 complexes with CD132 (common gamma chain) to form the IL-7 receptor. It is expressed by immature B cells, thymocytes, and most mature T cells, with transient down-regulation upon activation. IL-7 binding triggers Jak/STAT signaling pathways essential for lymphocyte development and T cell homeostasis.
CD127 expression is notably down-regulated in CD4+CD25+ regulatory T cells (Tregs), making it a useful marker to distinguish true Tregs enriched for Foxp3 expression and suppressive activity.
Binding of eBioRDR5 to PBMCs is blocked by recombinant human IL-7 (Product #14-1079-80).

Applications Tested: Pre-titrated and validated for flow cytometry of normal human peripheral blood cells.
Use at 5 µL (0.125 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Target Information
CD127 (IL-7Rα) is a glycoprotein involved in lymphopoiesis regulation. The active IL7R complex is an alpha/gamma heterodimer; the gamma chain activates signal transduction, while the alpha chain mediates specific signaling. CD127 promotes proliferation of precursor B cells, thymocytes, and mature T cells. Dysfunctions are associated with severe combined immunodeficiency disorders.
