Product Specific Information

The monoclonal antibody 87G recognizes human HLA-G, a nonclassical MHC molecule involved in immune inhibition.
HLA-G has seven isoforms, and 87G detects both HLA-G1 and soluble HLA-G5. Expression occurs mainly in fetal trophoblast cells, protecting the fetus from maternal immune attack.
HLA-G expression is also found in certain tumors (melanoma, breast carcinoma, renal carcinoma, CLL, AML, B-CLL), pancreatic islets, erythroid and endothelial progenitors, and thymic medulla.
HLA-G⁺ CD4⁺ or CD8⁺ cells act as regulatory cells with distinct cytokine profiles. Receptors include CD85j/ILT2, CD85d/ILT4, and CD158.
Recent studies suggest roles in transplant tolerance and immune regulation.

Applications Tested: Pre-titrated and tested using stimulated U937 cells at 5 µL (0.25 µg)/test.
A test corresponds to staining a cell sample (10⁵–10⁸ cells) in 100 µL final volume.

Target Information

HLA-G is a class I heavy chain paralogue forming a heterodimer with β2-microglobulin.
It is expressed on fetal-derived placental cells and consists of an ~45 kDa heavy chain encoded by 8 exons:

  • Exon 1: Leader peptide
  • Exons 2–3: α1 and α2 domains (peptide-binding)
  • Exon 4: α3 domain
  • Exon 5: Transmembrane region
  • Exon 6: Cytoplasmic tail

HLA and MHC antibodies are essential tools in Immunopeptidomics, enabling neoantigen identification and characterization via LC-MS/MS.
