Product Specific Information

The monoclonal antibody 3D12 (HLA-E) recognizes human HLA-E, a member of the Human Leukocyte Antigen family.
HLA-E is an MHC Class IB molecule involved in NK cell recognition. Binding of peptides derived from other Class I leader sequences to HLA-E and β2-microglobulin complex leads to interaction with NKG2A on NK cells, protecting HLA-G-expressing cells from NK cell-mediated killing. HLA-E expression is ubiquitous.

  • Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells
    • Recommended use: 5 µL (0.5 µg) per test
    • A test is defined as staining a cell sample in a final volume of 100 µL
    • Cell number: 10⁵–10⁸ cells/test (empirically determined)
  • Excitation: 488–561 nm
  • Emission: 578 nm

Target Information

HLA-E belongs to the HLA class I heavy chain paralogues. This molecule is a heterodimer composed of a heavy chain and a light chain (β2-microglobulin).
The heavy chain (~45 kDa) gene contains 8 exons:

  1. Exon 1: Leader peptide
  2. Exons 2–3: α1 and α2 domains (peptide-binding)
  3. Exon 4: α3 domain
  4. Exon 5: Transmembrane region
  5. Exons 6–7: Cytoplasmic tail

HLA-E binds a restricted subset of peptides derived from leader peptides of other class I molecules.
HLA and MHC antibodies are crucial in Immunopeptidomics, aiding identification and characterization of neoantigens via HPLC-MS/MS.
