This WT31 monoclonal antibody recognizes the human alpha beta T cell receptor (TCR) complexed with the CD3 epsilon chain. The TCR is composed of a 44-kDa alpha and a 37-kDa beta chain, with specificity determined by V, D, and J gene rearrangements. It associates with the CD3 complex to recognize MHC-bound antigens presented by antigen-presenting cells such as B cells, dendritic cells, and macrophages, leading to T cell activation and cytokine production.

The WT31 antibody has been reported to induce proliferation of resting T cells and inhibit cytotoxic T cell activity. Some anti-CD3 antibodies may partially inhibit WT31 staining; clone SK7 (anti-human CD3) is recommended for two-color experiments.

Target Information

The T cell antigen receptor (TCR) is a ligand-specific α/β heterodimer associated with invariant CD3 subunits (γ, δ, ε, ζ). This complex is essential for surface expression and signal transduction during T cell activation, leading to differentiation and cytokine production. Most T cells express the α/β TCR, while γ/δ T cells are localized mainly in epithelial tissues.

TCR recognition of self-peptides is crucial for immune tolerance; aberrant recognition is linked to autoimmune diseases and tumor-associated mutations. Superantigen engagement of TCRs can lead to toxic shock syndrome. Autoantibodies against TCR Vβ segments have been found in rheumatoid arthritis and systemic lupus erythematosus, potentially modulating inflammatory responses.
