Target Information

Amivantamab is a biosimilar that targets EGFR (Epidermal Growth Factor Receptor, HER1, ErbB1), encoded by the EGFR gene on chromosome 7 in humans.
EGFR belongs to the HER/ERbB family of receptor tyrosine kinases, including ERbB2, ERbB3, and ERbB4. Ligand binding (e.g., EGF, TGF-α) induces receptor dimerization and autophosphorylation, activating downstream signaling cascades such as Ras–ERK, PI3K–Akt, Jak–STAT, and PKC pathways that regulate cell growth and proliferation.
Phosphorylation at Y1086 enables GRB2 binding and MAPK pathway activation. Activated EGFR is endocytosed and either degraded or recycled.
Mutations in EGFR are linked to lung cancer and other malignancies, with overexpression observed in tumors of the head and neck, brain, bladder, stomach, breast, lung, endometrium, cervix, vulva, ovary, esophagus, and squamous cell carcinoma.
