Target Information
HIF1-alpha (HIF1A) is a subunit of HIF1, a transcription factor found in mammalian cells under reduced oxygen tension. HIF-1 is a heterodimer consisting of alpha and beta subunits, both belonging to the PAS family of transcription factors. It regulates the adaptive response to hypoxia by activating transcription of genes such as erythropoietin, glucose transporters, glycolytic enzymes, and VEGF.
HIF1-alpha controls hypoxia-mediated apoptosis, cell proliferation, and tumor angiogenesis. Under hypoxic conditions, HIF1-alpha interacts with p53 and undergoes MDM2-mediated ubiquitination and degradation. Induction of NOX4 by HIF1-alpha helps maintain ROS levels after hypoxia and promote proliferation. In humans, HIF1A is located on chromosome 14q. The C-terminal binds to p300, forming complexes that induce hypoxia-responsive genes.
Hypoxia contributes to major human diseases, including myocardial and cerebral ischemia, cancer, pulmonary hypertension, congenital heart disease, and chronic obstructive pulmonary disease.
