Target Information
Hypermethylated in cancer (HIC-1) was originally identified as a target of p53-induced gene expression.
HIC-1 is deleted in Miller-Dieker syndrome (MDS) and frequently suppressed in leukemia and various cancers due to hypermethylation of specific DNA regions, leading to transcriptional silencing.
These findings indicate that HIC-1 acts as a tumor suppressor protein mediating transcriptional repression.
HIC-1 is ubiquitously expressed in adult tissues and characterized by five zinc fingers and an N-terminal broad complex POZ (BTB) domain.
In BTB/POZ containing proteins such as BCL-6 and PLZF, this domain interacts with the SMRT/N-CoR-mSin3A HDAC complex to repress gene transcription.
Unlike these proteins, HIC-1 does not interact with HDAC components, suggesting its transcriptional repression is independent of the POZ/BTB domain.
