Product Specific Information

The eBio64DEC17 antibody reacts with human IL-17A and functions as a neutralizing antibody.
IL-17A is a CD4+ T cell-derived cytokine promoting inflammatory responses and is elevated in autoimmune and inflammatory diseases such as rheumatoid arthritis, asthma, multiple sclerosis, psoriasis, and transplant rejection.
Human IL-17A exists as glycosylated homodimers (20–30 kDa) and stimulates epithelial, endothelial, and fibroblastic cells to secrete IL-6, IL-8, G-CSF, and PGE2.
It also enhances ICAM-1 expression and supports hematopoietic progenitor proliferation and neutrophil maturation.

IL-23-dependent Th17 cells produce IL-17A, distinct from Th1/Th2 pathways. IFN-γ and IL-4 negatively regulate Th17 differentiation.
Intracellular staining by eBio64DEC17 identifies the same cell population as eBio64CAP17.
This antibody is tested for intracellular staining followed by flow cytometry using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00).

Recommended Use: 5 µL (0.5 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.

Fluorochrome Information:
Brilliant Ultra Violet™ 805 (BUV805) emits at 797 nm and is suitable for cytometers equipped with a 355 nm UV laser.
Use Super Bright Complete Staining Buffer (Product #SB-4401) or Brilliant Stain Buffer (Product #00-4409-75) to minimize non-specific polymer interactions.

Handling Notes:

• Light-sensitive; protect from light.
• Compatible with short-term methanol fixation (<1 hour).
• Store fixed samples at 4°C in dark for up to 3 days.
• Excitation: 355 nm; Emission: 797 nm; Laser: Ultraviolet.

Target Information

Interleukin-17A (IL-17A, CTLA-8) is a CD4+ T cell-derived cytokine that promotes inflammatory responses and is elevated in several autoimmune conditions.
IL-17A is a 32 kDa disulfide-linked homodimer (136 amino acids) and signals through IL-17R/CDw217.
It regulates NF-κB and MAPK pathways, stimulates IL-6 and COX-2 expression, and enhances nitric oxide production.
